The iPhone Dev Team has released updated versions of PwnageTool and QuickPwn for iPhone, iPhone 3G, and iPod touch. The update enables iPhone OS 2.2.1 to work properly with existing jailbreak exploits.
However, the team stresses that iPhone OS 2.2.1 contains an updated iPhone 3G baseband. The updated baseband cannot be downgraded, and does successfully block the yellowsn0w SIM unlock exploit.
The workaround to upgrade, and stay unlocked, is a bit complex. The iPhone 3G must not be updated via iTunes to 2.2.1. Instead, only use PwnageTool on a Mac to jailbreak the iPhone 3G up to 2.2.1. PwnageTool suppresses the baseband update by default, and ensures that the iPhone 3G has 2.2.1, but with 2.2.0’s baseband. Then, re-run the (updated) yellowsn0w exploit to leave you with an iPhone 3G on 2.2.1, and unlocked.

You certainly will be able to jailbreak with PwnageTool. From what I have seen, it should still work for unlocking as well. Apple really gave up on preventing the original iPhone from being unlocked, as opposed to iPhone 3G.
